Harry Maguire started the game playing from the right flank which saw him delivered a measured cross which led to the opener.
Maguire cross was met by Manuel Ugarte who flicked on for Casemiro to head home in the 30th minute.
The header saw United breaking the deadlock against a side that had not conceded at home in Europe since August.
Seven minutes later, centre-back Dani Vivian hauled down United striker Rasmus Højlund inside the box.
After a VAR review, Vivian received his marching orders, and Bruno Fernandes coolly dispatched the spot-kick to make it 2-0 in the 37th minute.
Just before the break, United’s talisman combined with Ugarte once more which saw them making it worst for the home team.
A clever backheel set up Fernandes, who slotted his second of the night in the 45th minute, sealing a dominant first-half display and leaving Athletic with an uphill task.
There was nothing much which happened in the remaining 45 minutes with both teams holding things down to finish 0-3.
With a three-goal cushion and the tie suspended until next Thursday at Old Trafford, Manchester United are heavy favourites to progress to the Europa League final.
Athletic Bilbao will need a near-miracle, attempting to overturn the deficit at a venue where United have been formidable this season.
